Hello everybody, and welcome to this NEJM Open Group Forum discussion on the article, “Inherited DNA-Repair Gene Mutations in Men with Metastatic Prostate Cancer.” My name is Nicholas and I will serve as a moderator for this thread for the duration of the discussion period. First off, I would like to congratulate Dr. Pritchard and colleagues on their novel study which characterized for the first time the frequency of inherited DNA-repair gene mutations in patients with metastatic prostate cancer and which identified mutations in 16 different DNA-repair genes associated with autosomal dominant syndromes predisposing for cancer. In addition, I would like to thank the experts who have generously given their time to provide insights as we delve more deeply into this important topic. As an introduction, this open forum is meant to encourage discussion and questions from individuals with differing levels of knowledge; I welcome all our guests to ask questions and encourage all our authors and experts to answer any questions that are posed.
